These surface proteins need to first be translocated across the bacterial cell membranes from the cytoplasm to the cell surface. Gram-negative bacteria have an additional periplasmic space, which Gram-positive bacteria lack, so they have a harder task of translocating proteins. The display of heterologous proteins on the bacterial cell surface normally requires the fusion of the protein with a surface protein, called a scaffold.
